Hi Joshua,
It depends on your intended meaning. As the lesson explains, you would use "une" to express "a/an" or "one", whereas you would use "la" to express "the".
So for example:
J'ai une guitare = I have a guitar / I have one guitar.
J'ai la guitare = I have the guitar.

This happens when the context admits a/an, i.e, indefinite article as opposed when the article 8s the which is definite.
