I got both quiz questions for this wrong because they were questions. There were no questions marks so one would assume that they were statements rather than questions - Lisez-vous ce livre is different to lisez-vous ce livre?

Yes, "Lisez-vous ce livre" is only possible as an inverted question. Hence the instruction for this test question to choose the correct punctuation: ! | . | ? | ...
Lisez-vous ce livre ? = Are you reading this book?
Attention: Don't confuse the inverted question with L'Impératif
Lisez ce livre = Read this book
